A: 5-HTP stands for 5-hydroxytryptophan. 5-HTP is classified as a dietary supplement; it is made from the seeds of an African plant, Griffonia simplicfolia. 5-HTP is a metabolite (a metabolic by-product) of the amino acid tryptophan, and it is converted by the body into serotonin, which acts in the brain to sooth a person's mind and comfort one from stress and worry. People are using 5-HTP for a variety of conditions, including weight loss, depression, anxiety, PMS, fibromyalgia, eating disorders, obsessive-compulsive disorder (OCD), and headaches. 5-hydroxytryptophan has not been evaluated by the FDA for safety, effectiveness, or purity. Establishment of maternal behavior: Successful reproduction in mammals demands that mothers become attached to and nourish their offspring immediately after birth. It is also important that non-lactating females do not manifest such nurturing behavior. The same events that affect the uterus and mammary gland at the time of birth also affect the brain. During parturition, there is an increase in concentration of oxytocin in cerebrospinal fluid, and oxytocin acting within the brain plays a major role in establishing maternal behavior.
In a landmark 1979 study3, Cort Pedersen and Arthur Prange at the University of North Carolina in Chapel Hill showed that giving oxytocin to virgin rats could trigger maternal behaviours: the animals would build nests, lick or crouch over unfamiliar pups and even return lost pups to the nest. Researchers went on to show that oxytocin signalling in the brains of prairie voles (Microtus ochrogaster) helps the animals to form lifelong pair bonds4 — a rarity among mammals. In 2012, researchers even found a version of oxytocin in the tiny roundworm Caenorhabditis elegans, where it helps the animals find and recognize mates5.
First developed in the 1980s by researchers at the University of Arizona, Melanotan is principally used for the treatment of skin disorders including vitiligo and erythropoietic protoporphyria that affect skin appearance and sensitivity (especially to sunlight). Thymosin beta-4 is a very large molecule. In fact, it is so large that it cannot fit entirely into the receptor. Different sections of the molecule have different activities. TB-500 is the part of thymosin beta-4 hormone which promotes the most useful effects (overall healing, repair, new blood and muscle cells). For medical applications it is more practical to use the TB-500 instead of the entire Thymosin Beta-4 protein.
Growth factors play an important role is enhancing structural repair of chronic wounds (Robson, 1997). KGF-2 (Robson et al., 2001), TGF-β (Robson et al., 1995), PDGF-BB (Mustoe et al., 1994; Kiritsy et al., 1995; Smiell et al., 1999), β-NGF (Muangman et al., 2004) have been shown to enhance re-epithelialization (Greenalgh, 1996 for review). The KGF-1 gene has been shown to improve cutaneous wound healing in a septic rat model when delivered in a plasmid (Lin et al., 2006). The PDGF-B gene carried in a plasmid mixed with a bovine collagen gel was reported to accelerate closure of patient diabetic ulcers (Mulder et al., 2009; Blume et al., 2011). KGF-2, PDGF-BB and FGF-L are commercially available as RepiferminTM, RegranexTM, and Trafermin to treat human chronic wounds. Data for the effects of PDGF-BB on back wounds of diabetic mice and for the effects of KGF-2 on chronic venous ulcers in patients is tabulated in Tables 10.3 and 10.4. Thymosin β4 accelerated keratinocyte migration in the wounds of old diabetic mice (Philp et al., 2003).
The uterine-contracting properties of the principle that would later be named oxytocin were discovered by British pharmacologist Sir Henry Hallett Dale in 1906, and its milk ejection property was described by Ott and Scott in 1910 and by Schafer and Mackenzie in 1911. In the 1920s, oxytocin and vasopressin were isolated from pituitary tissue and given their current names. The word oxytocin was coined from the term oxytocic, Greek ὀξύς, oxys, and τοκετός , toketos, meaning "quick birth".
Oxytocin in a nine amino acid peptide that is synthesized in hypothalamic neurons and transported down axons of the posterior pituitary for secretion into blood. Oxytocin is also secreted within the brain and from a few other tissues, including the ovaries and testes. Oxytocin differs from antidiuretic hormone in two of the nine amino acids. Both hormones are packaged into granules and secreted along with carrier proteins called neurophysins.
Ingroup bonding: Oxytocin can increase positive attitudes, such as bonding, toward individuals with similar characteristics, who then become classified as "in-group" members, whereas individuals who are dissimilar become classified as "out-group" members. Race can be used as an example of in-group and out-group tendencies because society often categorizes individuals into groups based on race (Caucasian, African American, Latino, etc.). One study that examined race and empathy found that participants receiving nasally administered oxytocin had stronger reactions to pictures of in-group members making pained faces than to pictures of out-group members with the same expression. This shows that oxytocin may be implicated in our ability to empathize with individuals of different races and could potentially translate into willingness to help individuals in pain or stressful situations. Moreover, individuals of one race may be more inclined to help individuals of the same race than individuals of another race when they are experiencing pain. Oxytocin has also been implicated in lying when lying would prove beneficial to other in-group members. In a study where such a relationship was examined, it was found that when individuals were administered oxytocin, rates of dishonesty in the participants' responses increased for their in-group members when a beneficial outcome for their group was expected. Both of these examples show the tendency of individuals to act in ways that benefit those considered to be members of their social group, or in-group.
I’m always interested in learning about better supplements for my health. I’ve heard a lot of good things about peptides, but this was the first time I’ve read about TB-500 in particular. It sounds like it can have a major impact on helping you recover from injuries, which is a huge deal in today’s world. Thymosin β4 has been tested in multicenter trials sponsored jointly by RegeneRx Biopharmaceuticals Inc (Rockville, MD, USA) and Sigma Tau (Pomezia, Italy) in the United States and Europe in patients with bed sores, ulcers caused by venostasis, and Epidermolysis bullosa simplex and was found to accelerate bed sore and stasis ulcer repair by one month. It has also been tested in patients with chronic neurotrophic corneal epithelial defects and found to promote repair.

One study investigating romantic stress that looked at nondepressed youth who went through a recent breakup and were given 60mg of Griffonica Simplicifonia (12.8mg 5-HTP) twice a day for 6 weeks in an open-label study noted reductions in percieved romantic stress when measured at the 3 week mark with no further improvement at 6 weeks; there was no control nor placebo group in this study.

Sit in a quiet area and practice the following: Take a slow, deep breath through your nose, allowing both your stomach and chest to rise. For now, Bartz isn’t sure why oxytocin can have such different effects. Her most educated guess is that the hormone triggers a biased trip down memory lane. Under its influence, people are more likely to remember information about their mother that fits with their current attitudes to relationships. If they are anxious, they’re more likely to remember the negative side of their early life. It’s a reasonable enough idea, and one that Bartz intends to test in the future. It will also be good to repeat the study in a larger group – 31 men make for a relatively small study.
Thymosin β4 was initially perceived as a thymic hormone. However this changed when it was discovered that it forms a 1:1 complex with G (globular) actin, and is present at high concentration in a wide range of mammalian cell types. When appropriate, G-actin monomers polymerize to form F (filamentous) actin, which, together with other proteins that bind to actin, comprise cellular microfilaments. Formation by G-actin of the complex with β-thymosin (= "sequestration") opposes this.
Bonding. In the Prairie Vole, oxytocin released into the brain of the female during sexual activity is important for forming a monogamous pair bond with her sexual partner. Vasopressin appears to have a similar effect in males. In people, plasma concentrations of oxytocin have been reported to be higher amongst people who claim to be falling in love. Oxytocin has a role in social behaviors in many species, and so it seems likely that it has similar roles in humans.6
I don’t recall any literature (research or anecdotal) suggesting that TB 500 showed efficacy with regard to repair of brain tissue. I have however read anecdotal comments regarding N-Acetyl Semax Amidate with regard to increase in BDNF (Brain-derived neurotrophic factor) and the growth of new neurons. I am currently testing N-Acetyl Semax Amidate for its nootropic properties however I have read post in which people suggest that it is “healing” brain damage.
That view has led some clinicians to try oxytocin as a treatment for psychiatric conditions such as autism spectrum disorder. But the early trials have had mixed results, and scientists are now seeking a deeper understanding of oxytocin and how it works in the brain. Researchers such as Froemke are showing that the hormone boosts neuronal signals in a way that could accentuate socially relevant input such as distress calls or possibly facial expressions. And clinical researchers are starting a wave of more ambitious trials to test whether oxytocin can help some types of autism.
Oxytocin was also correlated with the longevity of a relationship. Couples with the highest levels were the ones still together six months later. They were also more attuned to each other than the low-oxytocin couples when Feldman asked them to talk about a shared positive experience. The high-oxytocin couples finished each other’s sentences, laughed together and touched each other more often. Feldman says it’s still not clear whether oxytocin was responsible for the stability of the couple’s bond six months later or if couples who weren’t as connected failed to trigger the oxytocin system.
A 2002 review concluded that although the data evaluated suggests that 5-HTP is more effective than placebo in the treatment of depression, the evidence was insufficient to be conclusive due to a lack of clinical data meeting the rigorous standards of today. More and larger studies using current methodologies are needed to determine if 5-HTP is truly effective in treating depression. In small controlled trials 5-HTP has also been reported to augment the antidepressant efficacy of the antidepressant clomipramine.
